Reconstituting a vial,
step by step.

Reconstitution is the point where a stable lyophilized powder becomes a much less stable solution. The procedure below is the standard laboratory method — it takes about five minutes, and the order of the steps matters more than the speed.

Lyophilized material is redissolved by directing diluent against the vial wall rather than onto the cake itself.
  1. Let the vial reach room temperature

    Take it out of cold storage and give it 15–20 minutes. Opening a cold vial in a warm room condenses atmospheric moisture directly onto the material, which starts hydrolysis before you have touched it.

  2. Collect the cake at the bottom

    Lyophilized powder is light and often ends up on the vial walls or in the stopper after transit. Tap the vial down on the bench, or centrifuge briefly. None of it is lost material.

  3. Sanitise both surfaces

    Wipe the vial stopper and the diluent stopper with an alcohol pad and let both air dry fully. Introducing solvent through a wet stopper carries whatever was on it into the vial.

  4. Draw your diluent

    Use the volume specified by your own protocol and the diluent appropriate to the compound. The Certificate of Analysis states solubility characteristics for the lot; concentration is determined by your experimental design, not by us.

  5. Direct the stream against the glass

    Insert the needle at an angle and let the diluent run down the inside wall of the vial. Injecting forcefully onto the cake shears the material and causes foaming, which denatures peptide at the air–liquid interface.

  6. Swirl — never shake

    Rotate the vial gently until the solution is clear. Shaking generates foam, and foam means an enormous air–liquid interface. Full dissolution can take a few minutes; let it.

  7. Inspect before use

    The solution should be clear and free of visible particulates. Cloudiness or undissolved material means the compound has not fully dissolved or has come out of solution — note it against the lot and do not proceed.

  8. Aliquot immediately

    Divide into single-use volumes now, while you are still standing there. Every freeze–thaw cycle degrades the material, and aliquoting is the only way to guarantee no volume is thawed twice.

  9. Label with the parent lot number

    Each aliquot gets the compound name, the lot number from the original vial, the concentration, and the date. Without the lot number an aliquot has no traceable provenance.

That depends on the compound, the diluent, and your storage conditions. As a general handling rule, reconstituted peptide is held at 2–8°C for short-term use and frozen in aliquots for anything longer. Consult the Certificate of Analysis for the specific lot.
Almost certainly not. Lyophilized material is very light and redistributes during shipping. Tap the vial down firmly or centrifuge briefly to collect it before opening.
No. That is exactly what aliquoting exists to prevent. Each freeze–thaw cycle imposes mechanical and chemical stress and the damage accumulates. Discard unused thawed material.
Cloudiness indicates incomplete dissolution or that the compound has precipitated. Do not use it. Record the observation against the lot number and contact us with that lot number.
The Certificate of Analysis states the solubility characteristics determined for that lot. Diluent selection is part of your experimental design — we publish the analytical data and cannot specify a protocol for your work.
