Material that has been stored incorrectly is of unknown quality regardless of what the Certificate of Analysis says — the certificate describes the lot as tested, under the conditions stated on it. Everything below exists to keep those conditions true.

Lyophilized and reconstituted material occupy two completely different stability regimes.
  1. Inspect on arrival

    Check whether the cold pack is still cold and whether the vial and seal are intact. Note the condition against the lot number in your intake log before anything goes into storage.

  2. Log it before you store it

    Compound, lot number transcribed from the vial itself, date received, arrival condition, and storage location. This takes two minutes now and is impossible to reconstruct later.

  3. Store lyophilized material cold, dark and dry

    -20°C, desiccated, protected from light, in the original sealed vial. Water is the primary degradation reactant — desiccation is doing as much work here as the temperature.

  4. Keep it out of the freezer door

    The door is the least thermally stable location in any freezer and cycles every time it opens. Store toward the back where the temperature actually holds.

  5. Equilibrate before every opening

    Room temperature before the stopper is pierced, every single time. A cold vial opened in a warm room pulls humid air onto cold glass and condenses water directly onto the material.

  6. Hold reconstituted material at 2–8°C

    Solution is a fundamentally less stable state than powder. Refrigerate for short-term use and freeze aliquots for anything beyond that.

  7. Aliquot to eliminate freeze–thaw

    Single-use volumes, divided at the point of reconstitution. This is the highest-value habit in peptide handling and it costs about ninety seconds.

  8. Use low-bind labware at low concentrations

    At dilute concentrations a meaningful fraction of peptide adsorbs onto untreated plastic. It does not degrade — it simply leaves solution, which reads as unexplained loss of activity.

  9. Minimise open-air time

    Oxygen and light both drive oxidation, particularly in sequences containing methionine, cysteine, or tryptophan. Cap it and put it back.

Lyophilized material is considerably more robust in transit than reconstituted material, and a brief excursion is usually tolerable. Record the observation against the lot number and contact us with it — we would rather assess it than have you guess.
Colder storage is not harmful, but -20°C desiccated is sufficient for lyophilized peptide and -80°C offers little additional benefit for the added handling. Consistency matters more than the specific setpoint.
Visually, you frequently cannot — which is the core problem. A collapsed or discoloured cake, visible moisture, or a solution that will not clarify are all flags. Absent those, the reliable answer is analytical, not visual.
No. The expiry date assumes the storage conditions stated on the certificate held throughout. A documented excursion invalidates that assumption.
Frost-free units work by cycling through brief warming periods to prevent ice buildup, which subjects contents to repeated small thermal excursions. A manual-defrost unit is preferable for long-term storage.
